Product Description
Fungal metabolite with the property to inhibit intracellular protein transport. It is a reversible inhibitor of protein translocation from the endoplasmic reticulum (ER) to the Golgi apparatus. It blocks binding of ADP-ribosylation factor to the Golgi apparatus and inhibits GDP-GTP exchange. Studies show prolonged treatment with BFA induce cell apoptosis. Brefeldin A-mediated apoptosis has been observed in human tumor cells.
Brefeldin A working concentrations and length of treatment can vary depending on desired effect. Inhibiton of ER to Golgi trafficking was observed as low as 100 ng/ml and apoptosis was observed with prolonged treatment at 10 μg/ml.
Synonym: γ-4-Dihdyroxy-2-(6-hydroxy-1- heptenyl)-4-cyclopentanecrotonic
Formulation: Brefeldin A Solution is supplied as a 1000X in DMSO, which should be diluted to 1X in cell culture medium. It is recommended that cells are cultured with brefeldin A for ≤ 24 hours, as this can become toxic for cell viability.
Concentration: 5.0 mg/ml
Storage & Handling: Store the Brefeldin A Solution (1,000X) at 4°C. Completely thaw the solution before use, DMSO freezes at 4°C. We recommend aliquotting the reagent into smaller volumes to avoid repeated freeze-thawing
